The homeostatic clearance of neutrophils regulates the kinetics of hematopoietic stem/progenitor cells (HSPCs) in the bone marrow. We present a protocol for studying the kinetics of HSPCs in mice using adoptive neutrophil transfer. We describe steps for isolating neutrophils from the bone marrow and performing an intravenous injection. We detail procedures for bone marrow flushing, immunomagnetic selection, neutrophil labeling, and successful tail-vein injection.
