SalmonellaentericaserovarTyphimuriumisanimportantzoonoticgastrointestinalpathogenresponsibleforfoodbornediseaseworldwide.Itisasuccessfulentericpathogenbecauseithasdevelopedvirulencestrategiesallowingittosurviveinahighlyinflamedintestinalenvironmentexploitinginflammationtoovercomecolonizationresistanceprovidedbyintestinalmicrobiota.Inthisstudy,weusedpigletsfeaturinganintactmicrobiota,whichnaturallydevelopgastroenteritis,asmodelforsalmonellosis.WecomparedtheeffectsontheintestinalmicrobiotainducedbyawildtypeandanattenuatedS.Typhimuriuminordertoevaluatewhetherthemodificationsarecorrelatedwiththevirulenceofthestrain.ThisstudyshowedthatSalmonellaaltersmicrobiotainavirulence-dependentmanner.WefoundthatthewildtypeS.Typhimuriuminducedinflammationandareductionofspecificprotectingmicrobiotaspecies(SCFA-producingbacteria)normallyinvolvedinprovidingabarrieragainstpathogens.Boththeseeffectscouldcontributetoimpaircolonizationresistance,increasingthehostsusceptibilitytowildtypeS.Typhimuriumcolonization.Incontrast,theattenuatedS.Typhimurium,whichischaracterizedbyareducedabilitytocolonizetheintestine,andbyaverymildinflammatoryresponse,wasunabletosuccessfullysustaincompetitionwiththemicrobiota.
