A novel pH-sensitive PEGylated liposome formulation for tumor targeting was investigated. The pH-sensitive liposomes were obtained by including 6% mol/mol of stearoyl-PEG-poly-sulfadimethoxine-methacrylate and 0.25% of PEG2kDa-DSPE in the liposome bilayer. In vitro, at physiologic conditions, the liposomes showed “stealth” behavior while at pH 6.5, the liposomes underwent size increasing, aggregation and selective association with tumor cells. Doxorubicin loaded liposomes showed pH dependent drug release profiles: the drug release rate w at pH 6.5 was twice than at pH 7.4.
